Legend: In mitochondria, fatty acids are synthesized by type II fatty acid synthase located in the matrix using malonate as a precursor. Octanoic acid, which is a major fatty acid synthesized in mitochondria, is used for biosynthesis of lipoic acid that is bound to the H protein of the GDC complex and the E2 subunits of PDH, KGDH, and BCKADH complexes as a cofactor.
Abbreviations: BCKADH, branched chain a-keto acid dehydrogenase; ER, enoyl-ACP reductase; GDC, glycine decarboxylase; HAD, 3-hydroxyacyl-ACP dehydrase; HAS, holo-ACP synthase; KAR, 3-ketoacyl-ACP reductase; KAS, 3-ketoacyl-ACP synthase; KGDH, a-ketoglutarate dehydrogenase; LS, lipoic acid synthase; LT, lipoyltransferase; MAS, Malonyl-ACP synthase; MCAMT, malonyl-CoA: ACP malonyltransferase; MCS, malonyl-CoA synthase; PDH, pyruvate dehydrogenase.
